Pilot System solution for SMB
The AZURA Pilot SMB System A29501 is optimized for the separation of binary mixtures in gram to kilogram-scale. Its special emphasis is put on the continuous operation mode and high productivity. The system configuration consists of four Pump 2.1L and seven 8-port multiposition valves integrated into four AZURA Assistans ASM 2.1L. Our user friendly software PurityChrom® MCC and the required IT hardware are included too. This system is our standard Pilot SMB configuration and is optimized to bring success on a wide range of applications both the separation of smal molecules and the purification of proteins via capture SMB. A maximum pressure of 100 bar, eight columns and a flowrate of up to 500 ml/min allows you to do a separation on columns up to 50 mm ID.

Simulated moving bed chromatography (SMBC) is increasingly applied as a separation technique in the pharmaceutical industry, production of fine chemicals, and in the field of bioengineering.
SMB is a method in process chromatography that enables substance mixtures to be continuously separated and extracted in two fractions. Its efficiency is significantly higher than batch chromatography, through better utilization of the column stationary phase.
Although in principle limited to two fraction separations, each partial fraction can be separated into a further fraction by repeated use of the SMB process – down to binary substance mixtures.
Typically, the SMB process is used for two component mixtures, like chiral separations or mixtures with a “leading” or a “trailing” key product peak in the chromatogram.
Due to their modular design, AZURA SMB systems can also be configured for other multi-column processes. KNAUER offers SMBC systems for purification up to 1000 kg/year.

How AZURA® SMB works
System Configuration
The standard AZURA® SMB systems consists of four pumps and seven multi-position valves. The devices are arranged as follows:
• Three pumps (Extract, Raffinate, Eluent) are placed inside the SMB cycle.
• The feed pump is placed outside the SMB cycle.
• Four valves are placed at the pump outlets.
• Three valves are placed at the pump inlets. The feed pump inlet is not connected to a multiposition valve.
Due to this configuration, the SMB system is very flexible for many different separation methods. Additionally to the process stability AZURA® SMB systems are outperforming every other SMB system on the market regarding material and configuration flexibility as well usable pressure range.
Solutions for continuous purification of xylitol
Simulated moving bed chromatography (SMBC) was applied for the purification of xylitol from fermentation mash of a fed batch culture. This process enabled to purify xylitol with nearly 100 % purity and recovery. Thus, allowing large scale purification of xylitol from biological xylose-xylitol conversion process.

Software Solution to control and monitor AZURA multi column chromatography systems
PurityChrom® MCC is a special version of our purification software PurityChrom® and is optimized to be used with continuous chromatography systems e.g. SMBC systems. PurityChrom® provides a very user-friendly and clearly structured interface. The system visualization offers a graphical representation and allows easy handling even of complex flow processes. Furthermore each device which is displayed in the fluidic scheme can be manually controlled, giving the opportunity to optimize, change and adapt your conditions also during the run. The new PurityChrom MCC SMB parameter wizard helps you to generate new SMB methods and optimize your parameters while the process is running. With the integrated starting point calculator, you can easily generate you SMB method with the adsorption isotherms of your substances. There is also a possibility to pause your method during a run. The hold function provides you with complete control over your chromatography process. Solvent visualization calculates the consumption of solvent for the current run and prevents your column from running dry. PurityChrom® allows flexible control of different devices from KNAUER and other manufacturers.
