The ÄKTAxpress™ is a dedicated chromatography system for multi-step purification of tagged proteins and antibodies in a research or small production setting. The ÄKTAxpress™ TWIN is a fully integrated protein purification system that delivers totally automated, multi-step purification of His- and GST-tagged proteins for small scale research. The two-module system accepts multiple samples, offering the ability to purify eight samples in approximately 11 hours. The single module unit can do fewer samples in similar time.
Optimized protocols with a choice of up to four purification steps minimizes the need for chromatography expertise. Tag removal and maintenance procedures can be integrated into the purification protocols, eliminating manual interference during a run. Purification schemes for antibodies, single and double affinity-tagged proteins are supported. A four-step protocol may consist of affinity chromatography (AC), desalting (DS), ion exchange chromatography (IEX) and gel filtration (GF). Desalting in-between different purification steps are optional
The ÄKTAxpress™ Twin Purifier System produces achieves highest possible purity - optimized protocols with up to four steps will deliver tagged proteins of near 100% purity. Built-in cleaning procedures between MAb samples to minimize carry over. The Yield - the system is capable of purifying target proteins in yields varying from 1 to 50+ mg. Each ÄKTAxpress™ system (module) consists of a combined system and sample pump (0.5 - 65 ml/min), an air sensor for complete sample loading, a UV monitor (280 or 254 nm), a conductivity monitor and fraction collector for 96- or 24 deep-well microplates. All components are under the control of a specially designed version of UNICORN.
An import file allows easier entry of sample information and bar-code reader can be connected if preferred. Such a compact design gives the system a small footprint to occupy a minimum of space. The modular design allows for a high degree of throughput-flexibility. Up to 12 modules can be operated by one computer in parallel. Each module can run the same or different protocols, and each one can be started at different times if preferred. The system setup can grow with increasing throughput needs by adding modules.
