calibration. Any temperature—
including below-ambient temperatures—
may be held indefinitely. This feature allows refrigeration after
an unattended run.
- Parallel printer port
(optionally) allows the instrument to print
thermal profiles in real time and document programs in hard copy.
Additionally, an analog jack can output to a 0-5 volt chart
recorder.
- Power-failure protection will resume program after power
interruptions of up to twelve hours.

MJR PTC-200-The
MJ Research PTC-200 is a 96 well gradient thermocycler that
can be supplied with different 'alpha blocks' such
as a 96 well or a
dual 48-well block. The dual
48-well block allows two independent programs to be run at the same
time. The PTC-200 also uses the Hot Bonnetª to allow oil-free
operation. Precise Peltier-effect heat pumps --robustly designed for
repeated cycling-- deliver the full temperature range of 0-100C
electronically, in a compact instrument with a footprint that measures
24cm x 28cm x 27cm high. Speedy ramping rates of; up to 3.0¡/sec in
models that hold 0.2ml tubes or V-well plates; Unparalleled
temperature homogeneity: ±0.4C within 30 seconds of arrival @ 60C for
all standard models. Large memory & amiable software that allows

PTC200
This thermal cycler is a
wonderful product. Aside from being easy to use, those
equipped with the MJ research gradient block
extend capability. The gradient feature allows the temperature
to vary as much as 24 degrees from the left side of the block to the
right side. The means that you are able to have 12 different
temperatures on one block. This is useful for optimizing conditions
for new primer sets. All one has to do is create master mixes and
distribute among the twelve wells across the block. This is a huge
time saver and does the work of 12 pcr machines with just one
instrument.
This machine works well with many types of
tubes and does not squish tubes as does some other machines that we
have. This is most likely due to the deepness of the wells in the
block.

The last offering from MJ is the
PTC-200 DNA Engine. The sleek design and rugged chassis are
suggestive of an Italian sports car. Interchangeable sample blocks
and heat pump assemblies allow the user to change format easily
between runs while retaining NIST-traceable accuracy. The lab need
only buy the single control unit and supplement it with the desired
Alpha modules, such as dual, independently controlled blocks,
microplate format, or a slide adapter for up to 16 slides. Thermal
control is achieved by either block, in-sample probe, or calculated
methods. Heating rates up to 3°C/second reduce reaction times while
±0.4°C temperature homogeneity can be reached within 30 seconds at
90°C. The Hot Bonnet heated lid controls pressure and height to
accommodate various plasticwares. A Power Bonnet is available for
robotic systems. Additionally, a single cycler can be networked to
control up to 15 separate instruments. A PC can also control and
monitor cyclers remotely and export data to video screens or
databases in real time.
For high-throughput applications or busy
labs, the PTC-225 or 'Tetrad' DNA Engine Tetrad may be the way to go. This
allows up to eight separate blocks to run synchronous or independent
reactions. A compact footprint is maintained by housing the power
supply separately outside the chassis, good for tight spaces or
robotic applications. All of the features of the PTC-200 are present
in this model as well.
The PTC-100 cyclers incorporate a Peltier
heating element into a more economical model that can be configured
to accommodate plates, slides, or tubes. An optional heated lid is
available as well as slide griddle adapters for amplifying up to
four slides at a time. A large memory that holds up to 320 programs
means that even crowded labs have ample programming space.
For small labs or those that want an
auxiliary cycler, the PTC-150 MiniCycler series may be just the
ticket. Incorporation of the Peltier-Joule heat pump increases
cycling speed with ramping rates up to 2.4°C/second. The temperature
may be programmed to allow refrigeration after completion of the
reaction. The compact, lightweight unit can be easily transported to
different areas of the lab. Power failure protection resumes cycling
after temporary power loss up to 12 hours.
Two models are available for in situ
work. The PTC-200/225 models mentioned above allow use of the Twin
Tower block that can hold up to 16x2 slides. The PTC-100-16MS is a
dedicated configuration of the PTC-100 series, able to hold up to 16
double- or single-thickness slides. The sample block is made from
oxygen-free copper milled from a single ingot and then plated in
nickel for durability. Memory for 200 programs, power-failure
protection, printer port, and a two-year warranty are standard.

contains an onboard Pentium®-class computer that delivers an
advanced graphical interface-ideal for managing production-grade
thermal cycling. For those labs that value quick training, the
creation and editing of protocols can be accomplished rapidly and

Real-time displays of block, sample and lid temperatures provide

This device is
a large capacity thermal cycler specifically designed for
high-throughput amplifications.
The DNA Engine
Tetrad consists of four sample-block/heat-pump modules suitable for
amplifications in either 96- or 384-well format. The temperature
control is provided by Peltier-Joule heat pumps which samples
thermal profiles every 50 milliseconds. The temperature of the
heatings blocks can be easily modulated at a rate of 3.0°C/sec
within a range from –5.0°C to 105°C. We perform our amplifications
in 384-well plates, but as mentioned above, it is also possible to
use 96-well plates.
What is very
practical is the fact that the thermal blocks operate independently,
not only in terms of PCR protocols, but also because they can still
operate even if one or more of the blocks is out of order.
In summary,
the DNA Engine Tetrad is one of the very few high-throughput PCR
machines available on the market and I think it is a good one. It’s
compact design and modular structure facilitate performing large
scale experiments. I recommend this device to any researcher
planning to start large-scale genomics projects.
